Enhancing Cell Recovery and Debris Removal for Frozen Lymphocytes for Single Cell Multi-Omics

BioLegend manufactures CITE-Seq assay kits (TotalSeq). A co-developer of the method and Curiox Business Development Director (formally of BioLegend), Dr. Bertrand Yeung explains how extensive cell washing is critical to study success in removing background antibody signal. Yet, the additional washing steps can cause significant cell loss, particularly with samples containing very low starting cell numbers (50-100,000 cells).

BioLegend has found that Laminar Wash technology offers a significant improvement in processing time, while offering comparable proteogenomics data, versus centrifugation. Laminar Wash can process up to 96 samples in less than 15 minutes, whereas it could take anywhere from 30 to 45 minutes for only 10-20 samples. Thus, Laminar Wash technology is the superior choice for labs that desire high throughput sample processing.

Dr. Antonino Montalbano explains that the Laminar Wash system at ImmunAI can outperform the centrifuge at cell recovery, particularly for samples with very low starting numbers of cells. In multiplexing multiple samples utilizing TotalSeq cell hashing, Laminar Wash yields more singlet cells, while reducing both doublets and negatives (i.e., cells unstained with antibody). The two methods generate similar gene expression QC data and CITE-Seq results.
