Laminar Wash Technology Enhances Cell Retention and Produces Cleaner, Clearer Data
When dissociating cells from tumor-bearing tissues, tissue debris and enzyme-induced fragments are major factors that can affect the recovery of tumor-infiltrating lymphocytes (TILs). In more invasive and immunologically challenging models, such as mice inoculated with B16F10 melanoma cells, the limited recovery of TILs decreases the likelihood of accurate characterization. The results presented here, using an unchallenged mouse sample and Laminar Wash technology, demonstrate that laminar washing of 1,000,000 tumor-infiltrating cells increases viable cell counts and reduces debris. This cleaner sample preparation method improves lymphocyte recovery to 90.6%, compared to just 10.5% with traditional centrifuge-based washing methods.

 

Laminar Wash Technology Enhances Neutrophil and Monocyte Retention for Improved Differential Blood Counts
A comparison study using a 16-color flow cytometry panel focused on T cells was conducted with 100 µL of human whole blood using Laminar Wash technology. Whole blood was first lysed with ammonium chloride buffer and then centrifuged once to reduce volume. The samples were aliquoted for antibody incubation and subsequently washed using either the HT2100 system or centrifugation in polystyrene tubes.

Replacing the centrifuge with the HT2100 system in a single wash step provided clear resolution of rare T cell populations, such as MAIT cells, gamma-delta T cells, and regulatory T cells. The study also showed higher retention of neutrophils and monocytes, which was quantified through direct acquisition on a flow cytometer, minimizing cell perturbations.
