Standardizing PBMC Washing in Multi-User Flow Cytometry Labs Using Pluto ALPHA Automated Centrifuge-Free Workflows

Manual centrifugation-based PBMC washing introduces operator variability, inconsistent cell recovery, and workflow bottlenecks — especially in multi-user lab settings.

This poster presents a head-to-head comparison of conventional centrifugation and automated centrifuge-free washing using the Pluto ALPHA workstation across surface staining and FoxP3 intracellular staining workflows. Key findings include high correlation in immune cell population frequencies (R² ≈ 0.999), comparable stain index across lineage markers, and similar CD45⁺ cell retention — with significantly reduced hands-on time.
