Automated Antibody Cocktailing Reduces Operator- and Site-Dependent Variability in High-Parameter Flow Cytometry Panel Preparation

Manual antibody cocktail preparation introduces operator- and site-dependent variability that can affect QC consistency in clinical flow cytometry labs running standardized panels.

This poster evaluates the Pluto Antibody Cocktailing Wizard (ACW) across 9-, 10-, and 24-color panels — eliminating inter-operator variability (5/6 markers ≤5% CV), cutting hands-on time from 6:26 to 3:18–4:34 min, and improving cross-site vial-to-vial consistency (0.4% CV vs. 1.9% manual, r=0.997 concordance) — without changes to established staining protocols.

Clinical flow labs running standardized panels across multiple technologists face a hidden QC risk: manual cocktail prep. This poster shows how automating that single step — via ACW — closes the gap between operators and sites, cutting hands-on time nearly in half while holding staining performance steady across 9-, 10-, and 24-color panels.
