This poster demonstrates how Pluto Code enables centrifuge-free, on-deck sample preparation directly within liquid-handling workflows (OT-2, Hamilton STAR), executing C-FREE gentle-wash physics as programmable mix and wash primitives. In a PBMC surface staining workflow, Pluto Code significantly reduced debris versus manual centrifuge SOP (6.8% vs 9.2%, p<0.001), while subpopulation frequencies tracked closely with SOP across two independent immunophenotyping panels (R² = 0.997 and 0.994) and Stain Index was retained or improved for most surface markers. In murine Foxp3 intracellular staining, Pluto Code significantly improved both CD45+ leukocyte retention and regulatory T-cell (Treg) recovery versus centrifuge SOP (paired t-test, p<0.01).
